Abstract:

The following data collection comprises several experiments that use confocal fluorescence microscopy and smRNA FISH to investigate the transcriptional mechanism of olfactory receptors (ORs) expression in Ooceraea biroi antennae. Experiments were carried out in whole antennae in which olfactory sensory neurons were stained with custom RNA FISH probes. These data demonstrate that bidirectional transcription occurs leading to 1) inhibition of transcription of upstream ORs in a tandem array and 2) non-canonical readthrough producing non-translated transcripts from ORs downstream of the initiation site.

Each subdirectory contains individual antennae stained for distinct genes. These names correspond to the data included in our publication. The target of each probe is specified in the subdirectory name (U34, Chymotrypsin, Lnc4, T79-intergenic, 9E88, etc.). The numbers 488, 546, 594, 647 correspond to the Alexa fluorophores used for staining and imaging. Therefore, “U34-B3-546” indicates that U34 is in the red Alexa546 channel. All samples are age-matched to P14 (pupal day 14). Each TIFF image represents a distinct biological replicate of the same stain.
